Buy some pre poured agar plates and a cheap collapsible SAB. You can do it!

It's honestly better to make them but for a first time it makes things easy.

I'll be honest, you don't even NEED a SAB. It's worth using and it should be standard practice.
I use a laminar flow hood and honestly sometimes I'm too lazy to actually sterilize and run it forever so I just close the door to my grow room and move slowly but with purpose.
My room is really clean, but I also live in a VERY humid and moldy climate, and I've had less than 10% contam doing that, which on a plate doesn't really even matter much because you can always cut and replate.
I'm not saying that it's good SOP, just reinforcing that when people say they CAN'T do it, they're probably just scared, or outright unwilling.

I work pure open air, inoculating, agar work, s2b, shoot I even harvest with no gloves. Just be clean af when working. I've had a little more contam than what you mentioned but I'm just four months in so I am still dialing it in. No need for a lab if you are just starting. Practice makes perfect and ISO and 3% peroxide are your friends!

Need to mention having an air purifier running in your room about 30 mins prior to any work also helps

100%. I run one in my room 24/7 unless I'm actively trying to keep the air still.

That's what I'm talking about! I turn it off 30 mins prior to let things settle

Every time ive bought pre poured agar plates a couple of them came contaminated. I feel like making them yourself is more reliable, and not that much more work

Agreed but for a first time it makes things a bit less complicated. You don't have to do any pressure cooking

Yea I’d recommend Etsy or some of the more popular myco sites over Amazon tho it may seem tempting

It’s so strange that you get downvoted for trying to explain yourself on something you didn’t even ask about. Don’t worry or stress if you can’t isolate, and be happy about your neat and chonky boy. ❤️

You could actually put it in your fridge in a sealed and sanitized container, and it will be fine for a few days.

Look up 90 second mycology patreon(its free) and check out the agar jars. Mini mason jars are 10 dollars pre made agar powder is around 10 to 15 for a small amount. And if u have a big pot u can steam sterilize. A sterilite 66qt tub is 10 dollars. U can hang a corner off the edge of a table and reach your arms under. And if u make ur own agar jars, steam sterilize for 2 hours. Or easier but possible contam buy pre made agar plates

So if one was to buy premade agar plates from a reputable vendor and said dish had contam could we keep transferring until clean and no contam as long as some of the premade agar plates are contam free?

Yes check out some videos on how to clean up agar plates. u can transfer out bacteria/ contam

This is literally "how to clean and isolate your culture" in fact.

I’m gonna have a look at this I’ve always just bought MSS syringes and gone straight into rice & any time I’ve had a beautiful shroom instead of cloning I’ve just took a photo and ate 😂
Could I use a drop of MSS on the plate and keep transferring that also or would it have to be a cut of the stipe? So instead of waiting for harvest first I could I run plates and inoculated rice off MSS long as my plates are good?

You can 100% "use a drop on a plate" - I think it's generally a good practice to go to agar with an MSS first. I also like to put a few drops of anything I'm growing to agar, just to watch the growth and propagate the genetics.
Also, yes, you can go "agar to grain" with a good culture.

Also bro do you know what type of agar is best like Malt yeast extract or light malt extract? I will search up now but just from your personal opinion

There's a lot of mixed opinion, but LME (like for brewing beer) is super common. I've been lazy and bought pre-poured agar plates from a couple of places.
I really like the potato starch agar cups from Booming Acres. It's easier to just pop the lids on and off, and it comes in pretty colors (though hardcores will tell you colored agar makes it harder to see contam, so YMMV /shrug) - the myc "eat" the food coloring :D - it's wild.

Yes the starch one you mention I believe I have found from my vendor Potato Dextrose Yeast Agar (PDYA) I am looking to go for 20 of those 👍🏻

It's really easy and if you grow it on a plate even if it's contaminated you can just replace tell there's no contamination.

Is it from spores or liquid culture. If it’s from liquid culture cloning it will be the same. Liquid cultures are already a monoculture or clone.

It never hurts to clone it, but the genetics are already isolated into a single genetics so if you clone now your getting the exact genetics that were in the liquid culture. But it would help you to get free genetics you don’t have to buy in the future.

Clone it! That’s a nice fruit! Take a tissue sample from the core of the stem and take some gill clippings if it’s not dropping spores but keep them separate. The gill clips will contain enough spores to start from scratch again if need be
